Brother Bilaal, who describes himself as the actor's ex-personal assistant, made the shock accusation in an now viral interview with internet personality Tasha K.
Bilaal claims he once walked in on the Men In Black star, now 55, and Martin engaged in a sex act in Martin's dressing room.
'I opened the door to Duane’s dressing room and that’s when I see Duane having anal sex with Will,' Brother Bilaal said.

In the chat, Bilaal said during the latter years of their relationship, Smith was not his friend, but rather he was Smith's friend.
After Pinkett Smith spoke publicly about their marital estrangement, Smith followed up shortly after with his own comments on the situation.
He said he had the keys to Martin's dressing room when he discovered the two actors engaging in sexual activity. And I just haven't been able to break that promise.'
In the interview with Kotb, Pinkett Smith was asked about the longtime rumors swirling around the Hollywood couple.
Kotb said: 'You and Will probably have more rumors about you and talk about you than others: "They're in an open marriage, they're swingers, he's gay, she's gay" - I mean a million things were out there.
Kotb asked Pinkett Smith, 'What, if anything, is true and did hearing all that stuff bother you?'
Pinkett Smith denied all of the accusations at once: 'I would say that none of that's true.

Will Smith has been forced to deny that he is gay after rumours emerged he had a sexual relationship with Duane Martin.
The King Richard actor, 55, has made headlines in recent weeks when his wife Jada Pinkett Smith revealed the pair had in fact been separated for many years.
She claimed the night at the Oscars where he slapped Chris Rock after the comedian made fun of Jada was the first time in years he had referred to her as his wife.
Now Will has been forced to speak out again to deny that he was caught having sex with Fresh Prince of Bel-Air co-star Duane, who appeared in a handful of episodes in the 90s.
The actor responded after an interview with a man purporting to be Will’s former personal assistant, Brother Bilaal, went viral on TikTok.
In it, he claims he once walked in on the two men having a sexual encounter in the I, Robot star’s dressing room.
A spokesperson for Will responded to the claims in a statement to TMZ, saying the story was ‘completely fabricated’ and ‘unequivocally false.’
A source went on to state that the Oscar-winning actor is considering legal action over the remarks.
Duane, who was married to My Wife And Kids star Tisha Campbell before they divorced in 2020, has not addressed the claims.
Will’s marriage to Jada has been well publicised since the pair first began dating in 1995, after his first marriage to Sheree Zampino came to an end.
They went on to tie the knot in 1997 and welcomed children Willow and Jaden Smith together, but recently Jada announced they have been separated for years.
However she has since insisted that despite their split, they have no plans to divorce – and while they live in separate houses she claimed they were ‘working towards’ reconciling.
Days later Jada clarified that they were ‘ separated for seven years – until Oscar night,’ appearing to confirm they are a couple again.
The Oscar night in question is, of course, the 2022 Academy Awards where Will stormed the stage and slapped comedian Chris after he made a joke about Jada’s alopecia, warning him: ‘Keep my wife’s name out of your … mouth.’

Bilaal said in response that 'if a woman is used to something the size of a baby leg, and you come in with a pinky toe, there's nothing you're gonna do to please her ...
'I want to say sorry to my kids and my family for the heat that I've brought on all of us.'
He added: 'I've reached out to Chris and the message that came back is that he's not ready to talk, and when he is he will reach out. My behavior was unacceptable, and I'm here whenever you're ready to talk.'

Bilaal said that he had been working on a Hollywood set and ordered to keep 'eyes on Will' in the moments leading to him witnessing the actors having sexual relations.
